The blaze broke out just after 3pm local time (8pm GMT) on the top floor of a building located at 565 W. 144th Street and Broadway.
Dramatic footage shows large plumes of smoke coming from the building as well as the entire roof on fire.
At least 100 firefighters are at the scene trying to
put out the blaze.
Two have reportedly suffered non-life-threatening injuries.
The fire is believed to have spread to four alarms at the six-storey building.
No injuries have been reported as of yet and it is unclear how many people were inside the building when the blaze started.
An estimated 40 families live in the building, which also has retail stores on the first floor and apartments on floors two to six.
Roads around the area remain closed to traffic.
According to witnesses, huge black plumes of smoke could be spotted from Central Park, 80 blocks away and from across the Hudson River in New Jersey.
